SiriusT3 key features and benefits:

 

The SiriusT3 is the next generation physchem measurement system from Sirius Analytical. It is the successor to our world renowned and market leading GLpKa and ProfilerSGA systems, which over the last decade have been established as the industry standard for pKalogP/D and solubility determinations by automatedpotentiometric and spectroscopic titration.

The precise automated probe management, calibration and cleaning routines on the SiriusT3 remove the risk of sample carry-over and probe damage due to user error. Probes are automatically moved through calibration and cleaning stations prior to each experiment. Calibration data is logged in the system software, providing a clear and persistent record of system performance.

The highest throughput assays on the SiriusT3 can measure pKas in under 4 minutes, using only 3µl of 10mM DMSO stock solution, collecting around 40 to 50datapoints from pH2.0 to pH12.0. No other pKa measurement system can offer richly detailed measured pKa data at this speed and small sample requirement. Other systems which rely on the use of multiple pH buffers cannot resolve heavily overlapping pKas, something which the SiriusT3 can do with ease.

 

Module Details:

The SiriusT3 comprises 3 modules which can function independently.Systems are available in different configurations, ranging from full systems with all components to lower cost systems with fewer modules.

Titrator module, which features an automated moving arm with the assay probes attached – pH electrode, UV dip probe, overhead stirrer, temperature sensor and capillaries for reagent addition. All of these probes have been miniaturised to fit into1ml solution volumes. There is a row of buffer and wash positions used for automatically calibrating and cleaning the probes, including a flowing water wash which cleans the electrode with fresh water after each experiment. At the front of the Titrator module is the sample position, which is temperature controlled with a Peltier device (fully controlled by computer), and an additional turbidity sensing device. This is the position where the titration for pKalogP/D or solubility is performed.

Dispenser module, which houses the precision micro dispensers for adding water, solvents and acid/base titrants from the reagent bottles stored in the drawer below. The instrument can have up to 6 different cosolvents loaded simultaneously (Methanol, Ethanol, DMSOAcetonitrileDioxaneDMFTHF and others). Also contained within this module is a UV/Vis spectrometer and light source, which is connected to a fibre optic dip probe. This provides the same high quality UV/Vis spectroscopic functionality as our well established D-PAS system for the GLpKa.

Autoloader module, which has a worktable with four 48-position vial trays. The trays are standard footprint, and easily integrated with existing robotics systems.  It has a robotic arm which automatically picks up and move vials to the sample position of the Titrator module. At the front of the Autoloader module is a built-in ultrasonic bath, which can be automatically used to aid the dissolution of poorly soluble compounds.
